Target Information
VEGF (Vascular Endothelial Growth Factor)는 45 kDa의 이황화 결합된 호모다이머 글리코단백질로, 혈관신생(angiogenesis)에 관여하며 종양의 성장과 전이에 중요한 역할을 합니다. VEGF는 내피세포의 생존, 증식, 화학주성, 혈관 투과성 등을 촉진합니다.
VEGF 패밀리에는 VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, 그리고 PIGF가 포함되며, 이들은 angiogenesis의 세포 신호 조절에 핵심적인 역할을 합니다. 대부분의 종양은 혈관신생을 필요로 하며, VEGF는 가장 강력한 혈관신생성 사이토카인으로 알려져 있습니다.
VEGF는 단일 유전자의 대체 스플라이싱으로 생성된 5가지 isoform을 가지며, 이들은 주로 내피세포에 발현되는 두 가지 수용체 flt-1 (VEGFR-1)과 flk-1/KDR (VEGFR-2)에 결합합니다. FLT1의 대체 스플라이싱 형태인 sFLT1은 VEGF를 높은 친화도로 결합하여 혈관신생을 억제하는 역할을 합니다.
VEGF의 상승된 발현은 POEMS 증후군(Crow-Fukase syndrome)과 관련이 있으며, 이는 여러 장기에 영향을 미치는 희귀 질환입니다.
