Target Information

Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported within the cell through two interconnected cycles mediated by approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR).
The biosynthetic cycle involves MPR transport of newly synthesized lysosomal enzymes from the trans-Golgi network to late endosomes or early lysosomes, while the endocytic cycle moves extracellular lysosomal enzymes from the plasma membrane to early endosomes via clathrin-coated vesicles.
MPRs cycle between these compartments every ~3 hours, predominantly residing in late endosomes, with fewer in the trans-Golgi network and about 10% at the cell surface.
In addition to man6P binding, MPR also binds type II insulin-like growth factor (IGF-II) simultaneously. A soluble truncated form (~240 kDa) representing the extracellular domain circulates in serum and retains ligand-binding capability.
