Target Information

Prolyl hydroxylase 4 is an enzyme that modifies HIF‑alpha. Classic prolyl hydroxylases are found in the endoplasmic reticulum and modify collagen, whereas HIF is an intracellular protein with distinct hydroxylation sites.
HIF is a transcriptional complex critical for oxygen homeostasis. Prolyl hydroxylase is essential for cellular oxygen sensing.
Under normoxic conditions, prolyl hydroxylases hydroxylate specific prolyl residues in HIF‑alpha, leading to its degradation. Under hypoxia, hydroxylation is reduced, stabilizing HIF and promoting angiogenesis.
Prolyl hydroxylase 4 catalyzes the post‑translational formation of 4‑hydroxyproline in HIF‑alpha proteins and may function as a cellular oxygen sensor, targeting HIF for proteasomal degradation via the von Hippel–Lindau ubiquitylation complex.
