Background
This gene encodes a mitogen-responsive phosphoprotein expressed in normal ovarian epithelial cells but down-regulated or absent in ovarian carcinoma cell lines, suggesting a role as a tumor suppressor. The protein binds to SH3 domains of GRB2, an adaptor protein linking tyrosine kinase receptors to SOS (a guanine nucleotide exchange factor for Ras), via its C-terminal proline-rich sequences. It may modulate growth factor/Ras pathways by competing with SOS for GRB2 binding. Alternatively spliced transcript variants encoding different isoforms have been identified (RefSeq, Oct 2011).
