Product Specific Information
The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor of the B7 family. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells and is upregulated upon stimulation. It functions as a counter-receptor for CD28 and CTLA-4 on T cells, playing a key role in T-B cell crosstalk and immune response regulation.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells; use 5 µL (0.2 µg) per test (100 µL final volume).
Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for optimal results.
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates. Add 5 µL per sample to antibody cocktails before combining with cells.
NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information
CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation. It is expressed by antigen-presenting cells and mediates immune regulation through binding interactions with CD28 and CTLA-4. Dysregulation of CD86 is associated with conditions such as gallbladder squamous cell carcinoma and myocarditis. Alternative splicing produces multiple isoforms.
