Target Information
Sin1 was initially identified as the human homolog of S. pombe SIN1. It is identical to Mip1, which interacts with MEKK2, a member of the MAPK signaling network.
MAPKAP1 prevents MEKK2 activation and dimerization by forming a complex with inactive MEKK2, thereby blocking JNK1/2, ERK1/2, p38, and ERK5 MAPKs.
MAPKAP1 also participates in the TOR signaling pathway, controlling cell growth and proliferation in response to nutrients and growth factors. It maintains the TOR/rictor complex, enabling specific phosphorylation of Akt, linking PI3K signaling to nutrient-regulated TOR activity.
Multiple alternatively spliced isoforms of MAPKAP1 have been identified.
