Product Specific Information

The eBioBV13 (BV13) monoclonal antibody reacts with mouse VE-Cadherin (CD144). VE-Cadherin is a 120 kDa type II cadherin family member with five extracellular cadherin domains and an actin cytoskeleton anchor. It mediates homophilic adhesion between endothelial cells and is localized at adherens junctions. VE-Cadherin is essential for endothelial cell migration, survival, contact-dependent growth inhibition, and assembly into tubular structures. VE-Cadherin+CD45- cells may give rise to hematopoietic cells.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested with mouse bEnd.3 cells at ≤1.0 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize concentration empirically.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by a violet laser (405 nm), emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Buffer Recommendation:
Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation Guidance:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal brightness loss.

Target Information

VE-cadherin is a calcium-dependent cell-cell adhesion glycoprotein located on chromosome 16 within a six-cadherin cluster. It plays a key role in endothelial cell cohesion and junction organization. The protein consists of five extracellular cadherin repeats, a transmembrane region, and a conserved cytoplasmic tail. It functions as a classic cadherin mediating homophilic adhesion and may influence cancer-related loss of heterozygosity events. An alternative splice variant has been described, though the full-length sequence remains undetermined.
