Target Information
Paraoxonase-1 (PON1) is a calcium-dependent ester hydrolase associated with apoA-I in HDL and prevents LDL oxidation. Polymorphisms in the human PON1 gene correlate with coronary artery disease. PON1 activity is reduced in individuals prone to atherosclerosis such as those with diabetes, familial hypercholesterolemia, and renal disease.
PON2 is an intracellular protein protecting against oxidative damage. PON3, similar to PON1, differs in substrate specificity and is implicated in reducing coronary artery disease risk. Human PON3 shares three conserved cysteine residues with PON1, essential for its in vivo activity. PON1-based catalytic scavengers may offer protection against organophosphate intoxication.
