Product Specific Information
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells.
Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions. Commonly used as a phenotypic marker for mouse T cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse splenocytes at ≤1.0 µg/test (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test (empirical optimization recommended).
Fluorochrome: Brilliant Ultra Violet™ 737 (BUV737) — emits at 732 nm; use with UV (355 nm) laser-equipped cytometers.
Buffer Recommendations:
- Use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409) to minimize non-specific polymer interactions.
Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:
- IC Fixation Buffer (Product # 00-8222)
- 1-step Fix/Lyse Solution (Product # 00-5333)
Methanol Compatibility: Short-term fixation compatible; avoid long-term methanol exposure (>1 hour).

Target Information
The CD3 complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits encoded on chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs mediating signal transduction.
CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Abnormal CD3 expression may occur in autoimmune diseases. Defects in the CD3 gene are associated with CD3 immunodeficiency.
