Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It exhibits immunosuppressive properties and has been used in organ transplant rejection treatments. CD3 subunits (gamma, delta, epsilon) are essential for TCR complex assembly and expression, and CD3 is expressed by thymocytes and all mature T cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test in 100 µL final volume.

Brilliant Ultra Violet™ 737 (BUV737):

  • Tandem dye emitting at 732 nm
  • Compatible with UV (355 nm) laser-equipped cytometers
  • Sensitive to photo-induced oxidation (protect from light)

When using polymer dye-conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (SB-4401) or Brilliant Stain Buffer (00-4409) to minimize non-specific interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333).
BUV737 is compatible with short-term methanol fixation but should not be stored in methanol-containing buffers for more than one hour.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. CD3 subunits (γ, δ, ε, ζ) are immunoglobulin superfamily members encoded on chromosome 11. They interact with signal transduction molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes and thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with immunodeficiency.
