Target Information

ADRBK1 (GRK2) is a serine/threonine kinase that phosphorylates G protein-coupled receptors (GPCRs). GRK2 (83 kDa) is ubiquitously expressed in mammals and plays a key role in regulating GPCR signaling. Upon ligand binding, GPCRs are phosphorylated by GRKs, leading to receptor internalization via beta-arrestin-1 and activation of the Ras–Raf–ERK1/2 pathway. GRK2 activity is regulated by phosphorylation (PKC, Src, ERK1/2) and protein interactions. ERK-mediated phosphorylation at serine 670 inactivates GRK2 in a feedback mechanism.
