Target Information

Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the γ phosphate of ATP, onto an acceptor amino acid in a substrate protein. Through this mechanism, they mediate most signal transduction in eukaryotic cells, regulating:

• Cellular metabolism
• Transcription
• Cell cycle progression
• Cytoskeletal rearrangement and movement
• Apoptosis and differentiation

The protein kinase family includes over 500 gene products and is classified into 8 major groups based on catalytic domain sequence comparison. The AGC kinase group consists of 63 kinases, including:

• PKA & PKG (cyclic nucleotide-regulated)
• PKC (diacylglycerol/phospholipid-dependent)
• RAC/Akt (related to PKA and PKC)
• ARK (G protein-coupled receptor kinases)
• RSK (ribosomal protein S6 kinases)
