Aminoacyl-tRNA synthetases are enzymes that charge tRNAs with their corresponding amino acids. The HARS gene encodes a cytoplasmic enzyme of the class II aminoacyl-tRNA synthetases, responsible for synthesizing histidyl-tRNA, essential for incorporating histidine into proteins. This gene is arranged head-to-head with HARSL on chromosome 5, sharing a bidirectional promoter. The HARS protein is a frequent autoantibody target in polymyositis/dermatomyositis.
