Target Information

The munc-18 interacting protein (Mint) family consists of evolutionarily conserved adaptor proteins involved in membrane transport and organization. In mammals, three isoforms exist: Mint1, Mint2, and Mint3. While the N-terminal regions vary, the C-terminal regions are highly conserved and contain phosphotyrosine-binding (PTB) and PSD-95/DLG-A/ZO-1 (PDZ) domains that mediate protein interactions.

Mint1 and Mint2 are primarily expressed in the brain and interact with Munc18, a key component of synaptic vesicle fusion. Mint3, expressed ubiquitously but at lower levels in the brain and testis, does not interact with Munc18. Instead, Mint3 binds to the Alzheimer’s disease-related amyloid precursor protein (APP) through its PTB and PDZ domains, potentially linking APP to transport machinery and regulating its endocytosis and metabolism. Abnormal APP metabolism is associated with early-onset Alzheimer’s disease.
