Product Specific Information

  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating the cell surface expression of the TCR complex.
T cell activation through the antigen receptor (TCR) involves the cytoplasmic tails of CD3 gamma, delta, epsilon, and zeta subunits. These CD3 subunits are members of the immunoglobulin superfamily encoded by closely linked genes on human chromosome 11.

CD3 components have long cytoplasmic tails that interact with signal transduction molecules via a double tyrosine-based motif. CD3 may play a role in TCR-induced growth arrest, cell survival, and proliferation.
The CD3 antigen is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
