Target Information

The CD3 subunit complex is crucial in transducing antigen-recognition signals into the cytoplasm of T cells and regulating the surface expression of the TCR complex.
T cell activation through the TCR involves the cytoplasmic tails of CD3 gamma, delta, epsilon, and zeta subunits — structurally related members of the immunoglobulin superfamily encoded by genes on human chromosome 11.

CD3 subunits have long cytoplasmic tails that associate with signal transduction molecules, mediated by a double tyrosine-based motif. CD3 may play roles in TCR-induced growth arrest, cell survival, and proliferation.

The CD3 antigen is expressed on:

  • 68–82% of normal peripheral blood lymphocytes
  • 65–85% of thymocytes
  • Purkinje cells in the cerebellum

It is not expressed on B or NK cells. Decreased T lymphocyte percentages may occur in some autoimmune diseases.
Defects in CD3 genes are associated with CD3 immunodeficiency.
