Target Information

Guanylate cyclase-activating protein (GCAP1) is a Ca²⁺-binding protein that upregulates synthesis of cGMP in photoreceptors. Mammalian GCAPs share over 90% sequence similarity, consisting of 201–205 amino acids with three conserved Ca²⁺-binding sites. The GUCA1A gene (GCAP1) is expressed exclusively in the retina.
Mutations such as Tyr99→Cys and Pro50→Leu in GUCA1A are associated with autosomal dominant cone dystrophy and phenotypic variability ranging from mild photophobia to rod-cone dystrophy. These mutations can alter protein stability and susceptibility to protease degradation, potentially contributing to retinal cell mortality.
