Target Information
KLRB1 (Killer cell lectin like receptor subfamily B, member 1, CD161) protein is a type II membrane protein with an external C terminus. It is expressed by almost all NK cells and a subset of CD3⁺ T cells. The KLRB1 antigen forms a homodimeric cell surface protein (40–44 kDa) and plays an inhibitory role on NK cells.
Activation of KLRB1 stimulates acid sphingomyelinase/SMPD1 and increases intracellular ceramide levels, along with activation of AKT1/PKB and RPS6KA1/RSK1 kinases, promoting T cell proliferation by anti-CD3 stimulation. KLRB1 acts as a lectin binding to Gal-α(1,3)Gal and N-acetyllactosamine epitopes, and interacts with CLEC2D/LLT1 ligand to inhibit NK cell-mediated cytotoxicity and interferon-gamma secretion.
KLRB1 contains an extracellular domain with C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
