Product Specific Information

Description:
The LN3 monoclonal antibody reacts with human MHC class II molecule HLA-DR, expressed on antigen-presenting cells (APCs) such as B cells, monocytes, macrophages, dendritic cells, and activated T cells. HLA-DR is a heterodimeric transmembrane protein composed of alpha and beta subunits and plays an essential role in peptide presentation to CD4⁺ T lymphocytes.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
The LN3 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.0075 µg) per test (100 µL final volume).
Applicable cell count: 10⁵–10⁸ cells/test.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.

Blocking Buffer Information:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance.
Use 5 µL per sample in antibody cocktails or single-color controls for optimal results.

Technology:
NovaFluor conjugates are based on Phiton™ technology, utilizing nucleic acid dye structures that enable engineered fluorescent signatures with optimized spillover and spread characteristics.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).

• The alpha chain gene (DRA) contains 5 exons encoding leader peptide, extracellular domains, transmembrane region, and cytoplasmic tail.
• DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5.
• DRB1 alleles determine peptide-binding specificity and are routinely typed for transplantation purposes.
• HLA-DR is expressed mainly on APCs such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T cells.
• MHC class II molecules bind processed peptides and present them to T-helper cells, initiating immune responses.

HLA and MHC antibodies are critical in Immunopeptidomics, supporting neoantigen identification and characterization via LC-MS/MS.
