Ankyrins are a family of proteins that link integral membrane proteins to the spectrin-actin cytoskeleton, playing key roles in cell motility, activation, proliferation, contact, and maintenance of specialized membrane domains.
Multiple isoforms of ankyrin are expressed in a tissue-specific, developmentally regulated manner. Most ankyrins contain:

1. An amino-terminal domain with multiple ankyrin repeats
2. A central region with a conserved spectrin-binding domain
3. A carboxy-terminal regulatory domain subject to variation

Ankyrin 1, the prototype of this family, was first discovered in erythrocytes and later found in brain and muscles. Mutations in erythrocytic ankyrin 1 are associated with hereditary spherocytosis. Complex splicing patterns and muscle-specific truncated isoforms have been identified.
