Target Information

The protein tyrosine phosphatase PTPN22 (also known as PTP22, LYP, PEP, formerly PTPN8) is a genetic variant associated with increased risk of several autoimmune diseases including type 1 diabetes and rheumatoid arthritis.
A missense SNP in PTPN22 encodes a hematopoietic-specific phosphatase known as Lyp, with the risk allele found in approximately 17% of the general Caucasian population and 28% of those with rheumatoid arthritis.
This variant may disrupt the P1 proline-rich motif critical for interaction with the SH3 domain of CSK (cytoplasmic tyrosine kinase), potentially altering T-cell activation regulation.
The specific interaction between CSK and PTPN22 suggests that PTPN22 may function as an effector and/or regulator of CSK in T cells and other hematopoietic cells.
