Target Information
The pax genes are a family of transcription factors active in specific tissues during early embryonic development. Pax family members possess a DNA-binding domain encoded by the paired box, allowing them to regulate genetic programs.
Several pax genes are associated with developmental mutations:
- pax-3: Waardenburg syndrome
- pax-6: Aniridia
- pax-2: Wilms tumor
During embryogenesis, Pax-2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. Expression is downregulated as the tubular epithelium matures, but persists in undifferentiated epithelium of Wilms tumors, correlating with proliferation of poorly differentiated epithelial cells. Expression of the Wilms tumor suppressor protein WT1 coincides with downregulation of pax-2, suggesting WT1 directly represses pax-2 transcription.
