Target Information
The pax genes are a family of transcription factors active in specific tissues during early embryonic development. Pax family members possess a DNA-binding domain encoded by the paired box, allowing them to function as transcription factors that regulate genetic programs.
Several pax genes are associated with developmental mutations:
- pax-3: Waardenburg syndrome
- pax-6: Aniridia
- pax-2: Wilms tumor
During embryogenesis, Pax-2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. Pax-2 mRNA and protein levels decrease as the tubular epithelium matures. However, Pax-2 expression persists in the undifferentiated epithelium of Wilms tumors, correlating with the proliferation of poorly differentiated epithelial cells. Expression of the Wilms tumor suppressor protein WT1 coincides with down-regulation of Pax-2, suggesting that WT1 directly represses Pax-2 transcription.
