Target Information
The munc-18 interacting protein (Mint) family is a group of evolutionarily conserved adaptor proteins involved in membrane transport and organization. Mammals have three Mint isoforms: Mint1, Mint2, and Mint3.
- Mint1 and Mint2 are expressed primarily in the brain and bind to Munc18, a key component of the synaptic vesicle fusion machinery.
- Mint3 is ubiquitously expressed at low levels in the brain and testis and does not interact with Munc18.
- Mint3 interacts with the Alzheimer’s disease-related amyloid precursor protein (APP) through its PTB and PDZ domains, linking APP to transport machinery components and regulating its transport, endocytosis, and metabolism.
Abnormal APP metabolism is associated with early-onset Alzheimer’s disease.
