Target Information

Tomuzotuximab은 EGFR을 표적으로 하는 바이오시밀러 항체입니다.
EGFR(Epidermal Growth Factor Receptor, HER1, ErbB1)은 인간 7번 염색체에 위치한 EGFR 유전자에 의해 인코딩되며, HER/ErbB 단백질 패밀리(ERbB2, ERbB3, ERbB4 포함)에 속합니다.
EGFR은 막관통 수용체로, EGF나 TGF-α 등의 리간드 결합 시 이합체화 및 세포질 도메인 내 티로신 잔기 자가인산화가 일어납니다. Src 비수용체 키나아제에 의해 추가 인산화되기도 하며, Ras–ERK, PI3K–Akt, Jak–STAT, PKC 등의 신호 경로를 활성화하여 세포 성장과 증식을 유도합니다.
특히 Y1086 잔기 인산화는 GRB2 어댑터 단백질 결합을 유도하여 MAPK 경로를 활성화합니다. 활성화 후 EGFR은 엔도시토시스되어 분해 또는 재활용됩니다.
EGFR 유전자 변이는 폐암 등과 관련이 있으며, 다양한 스플라이싱 변이체가 존재합니다. EGFR의 과발현은 여러 종양에서 불량한 예후와 관련 있으며, 두경부, 뇌, 방광, 위, 유방, 폐, 자궁내막, 자궁경부, 외음부, 난소, 식도, 위 및 편평세포암 등에서 관찰됩니다.
