Target Information
The WRN gene encodes a member of the RecQ and DEAH subfamilies of DNA and RNA helicases. These enzymes are essential in DNA metabolism, including transcription, replication, recombination, and repair. The WRN protein contains a nuclear localization signal at the C-terminus and predominantly localizes to the nucleolus. It exhibits 3’ to 5’ DNA helicase and exonuclease activities. Interaction with the Ku70/80 heterodimer suggests a role in double-strand DNA break repair. Mutations in WRN cause Werner syndrome, an autosomal recessive disorder characterized by premature aging.
