Target Information
BRCA1 (breast and ovarian cancer susceptibility protein 1)은 핵 인산화 단백질로서, 유전체 안정성을 유지하고 종양 억제 단백질, DNA 손상 감지 인자, 신호 전달 인자들과 함께 BASC(BRCA1 associated genome surveillance complex)라는 다단위 복합체를 형성하여 종양 억제 역할을 합니다.
BRCA1은 RNA polymerase II와 결합하며, C-말단 도메인을 통해 histone deacetylase 복합체와도 상호작용합니다. 이 단백질은 전사, 이중 가닥 DNA 절단 복구, 재조합에 관여합니다.
BRCA1 유전자 돌연변이는 유전성 유방암의 약 40%, 유전성 유방 및 난소암의 80% 이상과 관련이 있습니다. 다양한 스플라이싱 변이체가 존재하며, 일부는 질병 관련 변이로 보고되었습니다. 관련된 유사유전자(pseudogene)도 염색체 17에 위치합니다.
