Product Specific Information
The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune regulation. PD-1 is expressed mainly on activated T and B lymphocytes. Its ligands, PD-L1 (B7-H1) and PD-L2 (B7-DC), show overlapping functions and are expressed on normal tissues and activated antigen-presenting cells.
Costaining experiments indicate that eBioJ105 recognizes a different epitope than MIH4 (Product # 11-9969-42).
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry using stimulated normal human peripheral blood cells. Use at 5 µL (1.0 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended detection filter: 710/50 bandpass. Ensure instrument compatibility.
Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Light Sensitivity:
Protect from light to prevent photo-induced oxidation.
Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C (dark conditions) with minimal effect on brightness and FRET efficiency.

Target Information
T-cell activation is regulated by inhibitory receptors such as PD-1 (PDCD-1), which binds ligands PD-L1 and PD-L2 on antigen-presenting cells (APCs) to suppress immune activation. PD-1 signaling limits immune responses in the absence of danger signals (e.g., LPS). PD-1 knockout mice exhibit hyperactivated immune systems and autoimmune symptoms. Although PD-1 has a predicted molecular weight of 55 kDa, it often migrates at a higher molecular weight in SDS-PAGE.
