The eB-h209 monoclonal antibody reacts with human CD209 (DC-SIGN), a 44 kDa type II transmembrane protein containing a C-type lectin binding domain. It binds ICAM-3, ICAM-2, and HIV virus. DC-SIGN is mainly expressed on human dendritic cells and functions as a receptor for capture and transmission of HIV to T cells, supporting the immune response.
eB-h209 was developed against a C-terminal peptide of human DC-SIGN.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on human monocyte-derived dendritic cells at 5 µL (0.06 µg)/test.
A test is defined as the antibody amount required to stain a cell sample in a final volume of 100 µL (typically 10⁵–10⁸ cells/test).

Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness loss.
Excitation/Emission: Excitation 488–561 nm; Emission 775 nm; compatible with Blue, Green, and Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The CD209 gene encodes a transmembrane receptor, commonly known as DC-SIGN, expressed on dendritic cells and macrophages. It plays a role in innate immunity and recognizes diverse pathogens including viruses and parasites.
The protein comprises an N-terminal transmembrane domain, a tandem-repeat neck domain, and a C-type lectin domain. The extracellular region mediates pathogen recognition and cell adhesion via carbohydrate ligand binding.
The neck region facilitates homo-oligomerization for high-avidity multivalent ligand binding. Variations in its 23 amino acid repeats affect ligand binding.
CD209 is closely related to L-SIGN (GeneID 10332), differing in ligand-binding and distribution. Alternative splicing produces multiple variants.
