Target Information
Lipolytic enzymes are essential for mobilizing fatty acids from triglyceride stores in adipose tissue. Dysfunctional lipolysis affects energy homeostasis and may contribute to obesity and insulin resistance.
Previously, hormone-sensitive lipase (HSL) was the only known enzyme hydrolyzing triglycerides in mammalian adipose tissue. It is now recognized that adipose triglyceride lipase (ATGL) catalyzes the initial step in triglyceride hydrolysis.
ATGL is highly expressed in adipose tissue of mice and humans, exhibits high substrate specificity for triacylglycerol, and associates with lipid droplets. Inhibition of ATGL significantly reduces total adipose acyl-hydrolase activity. Thus, ATGL and HSL coordinately catabolize stored triglycerides in adipose tissue of mammals.
