Product Specific Information

The monoclonal antibody SolA15 recognizes mouse and rat Ki-67, a 300 kDa nuclear protein expressed during all active phases of the cell cycle (G1, S, G2, and mitosis), but absent in resting cells (G0). Ki-67 redistributes from the nucleus to chromosomes during mitosis and serves as a marker for determining the growth fraction of cell populations.

SolA15 also reacts with human, non-human primate, and canine Ki-67. It is reported for use in intracellular staining followed by flow cytometric analysis.

Recommended use:

  • Tested with Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00)
  • ≤ 0.25 µg per test (100 µL final volume)
  • Cell number range: 10⁵–10⁸ cells/test
  • Carefully titrate for optimal performance

Fluorochrome information:
Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm, compatible with UV (355 nm) laser-equipped cytometers.

Staining buffer recommendations:
Use Super Bright Complete Staining Buffer (Product #SB-4401) or Brilliant Stain Buffer (Product #00-4409-75) to minimize polymer dye interactions.

Light sensitivity:
Protect vial and samples from light; the dye is sensitive to photo-induced oxidation.

Fixation compatibility:
Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) with minimal brightness loss.
BUV737 is compatible with short-term methanol fixation (<1 hour).

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin and heterochromatin regions and shows cell cycle-dependent localization. Ki-67 serves as a proliferation marker, absent in G0 cells and increasing through mitosis.

In neoplastic tissues, the Ki-67 labeling index correlates with tumor grade and prognosis. It is widely used as a neuronal and proliferation marker in research.
