Product Specific Information

The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4).
IRF4 is a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.

  • In B cells, IRF4 is highly expressed in mature plasma cells and regulates differentiation.
  • In T cells, IRF4 is involved in Treg, Th2, Th9, and Th17 cell development and function.
  • IRF4 expression is upregulated upon T cell activation and induces Blimp1 expression and IL-10 production.
  • It also plays roles in macrophage polarization and is upregulated in various hematologic cancers.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis.

Applications Tested:

  • Tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells using the Foxp3/Transcription Factor Buffer Set (Product # 00-5523-00) and protocol.
    Refer to BestProtocols® Section (Protocol B: One-step protocol for intracellular (nuclear) proteins).
    Use ≤ 0.5 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Fluorophore Information:

  • PerCP-eFluor® 710 emits at 710 nm and is excited at 488 nm.
  • Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
  • Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

The IRF (interferon regulatory factor) family of transcription factors is characterized by a unique tryptophan pentad repeat DNA-binding domain.
IRFs regulate interferon responses to viral infection and interferon-inducible genes.
IRF4 is lymphocyte-specific and negatively regulates TLR signaling, essential for innate and adaptive immune activation.
A chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH may cause multiple myeloma.
Alternatively spliced transcript variants exist.
