Target Information

DNA-binding protein SATB2 binds to DNA at the nuclear matrix- or scaffold-associated regions. It recognizes the sugar-phosphate structure of double-stranded DNA and functions as a transcription factor controlling nuclear gene expression by binding to matrix attachment regions (MARs) of DNA and inducing local chromatin-loop remodeling. SATB2 also acts as a docking site for chromatin remodeling enzymes, recruiting corepressors (HDACs) or coactivators (HATs) to promoters and enhancers.

SATB2 is required for the initiation of upper-layer neuron (UL1) specific genetic programs and for the inactivation of deep-layer neuron (DL) and UL2 specific genes, likely through modulation of BCL11B expression. It acts as a repressor of Ctip2 and serves as a regulatory determinant of corticocortical connections in the developing cerebral cortex. Mutations in the SATB2 gene are associated with isolated cleft palate (CPI).
