Target Information
Beta-catenin은 세포 간 부착 구조(adherens junction, AJ)의 구성 요소로 처음 확인된 단백질입니다. AJ는 상피세포층의 형성과 유지에 필수적이며, 세포 성장과 부착을 조절합니다.
Beta-catenin은 E-cadherin의 세포질 도메인과 결합하여 E-cadherin을 alpha-catenin에 연결하고, 이는 다시 actin 세포골격에 부착을 매개합니다.
또한, Beta-catenin은 adenomatous polyposis coli 단백질 및 미세소관을 포함하는 세포골격 복합체와 상호작용하며, tyrosine kinase, phosphatase, Wnt/Wingless 등 다양한 신호전달 경로에 관여합니다.
이러한 상호작용은 형태형성(morphogenesis)을 조절할 수 있습니다. Beta-catenin은 모낭세포, 기저세포, 외·내근초 세포 등에서 발현되며, pilomatrixoma, medulloblastoma, colorectal cancer, ovarian cancer 등 여러 종양과 관련이 있습니다.
핵 내에서는 Lef/Tcf 전사인자와 함께 작용하여 cyclin D, c-myc 등의 유전자 발현을 촉진함으로써 세포 증식을 유도합니다. 이러한 기능은 종양 발생과 관련된 분자적 기반을 제공합니다.
