Product Specific Information

The M1/69 monoclonal antibody recognizes the mouse CD24 molecule, also known as Heat Stable Antigen (HSA). This 35–50 kDa GPI-anchored protein is expressed on erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. CD24 expression varies due to glycosylation differences and is used to distinguish B-cell development stages in mouse bone marrow. CD24 also interacts with P-selectin (CD62P).

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes and thymocytes at ≤ 0.8 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize concentration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, add 5 µL per 100 µL of cell sample.

Target Information

CD24 is a GPI-anchored glycoprotein expressed during multiple stages of B-cell development, from pro-B cells in bone marrow to mature B cells. Expression is low or absent in plasma cells. CD24 is commonly found in B-lineage acute lymphoblastic leukemia, B-cell CCLs, and non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity control.
