Product Specific Information

The M1/69 monoclonal antibody reacts with mouse CD24 (Heat Stable Antigen, HSA), a 35–50 kDa molecule anchored via phosphatidylinositol. It is expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. CD24 is variably glycosylated, resulting in heterogeneity of molecular mass across lineages. Expression of CD24 helps resolve stages of B lymphopoiesis in mouse bone marrow. P-selectin (CD62P) has been reported to bind to CD24.

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes and thymocytes; ≤ 0.8 µg/test (100 µL final volume).
Cell number per test: 10⁵–10⁸ cells (empirical determination recommended).
Careful titration recommended for optimal assay performance.

NovaFluor Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample before combining with cells.
For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Technology:
Based on Phiton™ technology using nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.
Excitation: 509 nm; Emission: 614 nm; Laser: 488 nm (Blue).

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development—from pro-B cells in bone marrow to mature B-cells. Expression is low or absent in plasma cells. CD24 is also found in B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity.
