Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.

Ki-67 expression varies by cell cycle phase:

• G1 phase: Perinucleolar expression
• G2 phase: Nuclear matrix expression
• M phase: Chromosomal expression

Because Ki-67 is absent in G0 cells and increases steadily through mitosis, it serves as a reliable proliferation marker. Ki-67 antibodies are commonly used to determine the proliferative fraction in neoplasms, showing strong correlation between low Ki-67 index and histologically low-grade tumors. It is also used as a neuronal marker for cell cycling and proliferation.
