Target Information

Mannose (or mannan)-binding lectin (MBL), also known as serum mannose-binding protein (MBP), initiates the lectin branch of the innate immune response by binding to the surface of pathogenic microorganisms and triggering complement fixation through an N-terminal collagen-like domain.
MBL plays a central role in immune defense by enabling antibody-independent neutralization of invading microbes. Mutations in human MBL are linked to immunodeficiency due to impaired complement activation.

In humans, MBL-associated serine proteases (MASP-1, MASP-2, MASP-3) and a truncated form of MASP-2 (sMAP or MAp19) form complexes with MBL to activate the lectin pathway of the complement system. Activated MASPs cleave downstream complement components. MASP-3, an alternatively spliced product of the MASP-1 gene, may inhibit MASP-2 by competing for MBL binding, thereby modulating complement activation.
