Target Information
The TEK receptor tyrosine kinase (TIE2) is expressed almost exclusively in endothelial cells. TEK signaling is critical for endothelial cell–smooth muscle cell communication in venous morphogenesis. Defects in TEK are associated with inherited venous malformations. The ligand angiopoietin-1 binds to this receptor and mediates signaling involved in embryonic vascular development. TIE2 expression is increased by thyroid-stimulating hormone and agents that elevate intracellular cAMP. Hematopoietic stem cells (HSCs) expressing TIE2 are quiescent, antiapoptotic, and adhere to osteoblasts in the bone marrow niche.
