Product Specific Information

The XMG1.2 antibody reacts with mouse interferon (IFN) gamma and functions as a neutralizing antibody. Mouse IFN gamma is a 20 kDa cytokine produced by activated T, B, and NK cells with antiviral and antiparasitic properties. It synergizes with TNF alpha to inhibit proliferation of normal and transformed cells and modulates immune responses across various cell types.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis.

Applications Tested:

  • Tested on stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
  • Recommended ≤ 0.5 µg per test (100 µL final volume).
  • Optimize antibody titration empirically.

Fluorophore Information:
Brilliant Ultra Violet™ 615 (BUV615) emits at 615 nm and requires a cytometer equipped with a 355 nm UV laser. Protect from light due to photo-induced oxidation sensitivity.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or Fix/Lyse Solution (Product #00-5333-54) with minimal impact on brightness.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor produced primarily by T-lymphocytes and NK cells. It is a dimeric glycoprotein (~45 kDa) that exerts antiviral and antiproliferative effects, induces MHC expression, activates macrophages, and enhances B cell immunoglobulin secretion.
Human IFN gamma does not cross-react with mouse IFN gamma. Its biological activity is species-specific and regulated by cytokines such as IL-2, FGF, and EGF, and downregulated by vitamin D3 or DMN. Mutations are associated with aplastic anemia.
