Target Information
AFAP1L2, also known as XB130, is structurally similar to actin-filament-associated protein (AFAP). It contains multiple SH2- and SH3-binding motifs, two pleckstrin homology domains, a coiled-coil region, and several phosphorylation sites. AFAP1L2 interacts with and is phosphorylated by c-Src tyrosine kinase. Knockdown of AFAP1L2 via siRNA reduces Src activity, IL-8 production, and EGF-induced phosphorylation of Akt and GSK3β, affecting cell cycle regulation in human lung epithelial cells. AFAP1L2 is highly expressed in thyroid tissue and serves as a substrate for RET/PTC kinase, implicated in papillary thyroid cancer. Down-regulation of AFAP1L2 reduces Akt activity, inhibiting cancer cell proliferation and survival, suggesting its potential as a therapeutic target. At least four isoforms of AFAP1L2 are known.
