Target Information
Cullin 4B (CUL4B)는 여러 cullin-RING 기반 E3 ubiquitin-protein ligase 복합체의 핵심 구성 요소로, 표적 단백질의 ubiquitination 및 proteasomal degradation을 매개합니다. 이 복합체의 기능적 특이성은 기질 인식 서브유닛에 따라 달라집니다.
CUL4B는 복합체 내에서 scaffold 단백질로 작용하여 기질과 ubiquitin-conjugating enzyme의 위치를 조정함으로써 촉매 반응에 기여합니다.
방사선 유도 DNA 손상에 대한 반응으로 CDT1, histone H2A, H3, H4의 polyubiquitination에 관여하며, DDB2에 의해 UV 손상된 크로마틴으로 표적화되어 DNA 수리 및 복제에 중요합니다.
Cyclin E의 ubiquitination을 통해 정상적인 G1 세포 주기 진행을 조절하며, 세포 성장 및 대사 조절과 관련된 mTOR 경로에도 영향을 미칩니다.
CUL4B의 mTORC1 매개 경로 조절은 26S proteasome 기능에 의존하며 MLST8과의 상호작용이 필요합니다.
