Product Specific Information

The 424LTS monoclonal antibody reacts with and blocks IL-17A binding to human interleukin 17 receptor A (IL-17RA, IL-17R). IL-17RA is a type I membrane protein (~96 kDa, 866 amino acids) involved in inflammatory responses to IL-17A, IL-17F, and IL-17AF heterodimer. It forms heterodimeric complexes with other receptor family members, including IL-17RC for IL-17A and IL-17F signaling, and IL-17RC for IL-17E (IL-25) signaling. IL-17RA expression is ubiquitous, with elevated levels on granulocytes and monocytes.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.5 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (alternative: 695/40 bandpass). Ensure instrument compatibility.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

Interleukin 17A (IL-17A) is a proinflammatory cytokine secreted by activated T-lymphocytes. It induces maturation of CD34-positive hematopoietic precursors into neutrophils. IL-17RA binds IL-17A and IL-17F with low affinity and plays a pathogenic role in inflammatory and autoimmune diseases such as rheumatoid arthritis. Defects in IL-17RA cause familial candidiasis type 5 (CANDF5), a rare disorder with impaired fungal clearance.
