Product Specific Information

This 8CH monoclonal antibody reacts with human IL-8 (CXCL8), a pro-inflammatory CXC chemokine. IL-8 is synthesized as a 99 amino acid precursor protein processed into several isoforms, with IL-8(72) and IL-8(77) being most common. IL-8(77) is mainly secreted by endothelial cells and mediates angiogenesis during fetal development, while IL-8(72) is predominant in adults, expressed by monocytes, macrophages, epithelial cells, and fibroblasts in response to inflammation and stress.

IL-8 activates and recruits neutrophils and influences T cell migration via CXCR1 and CXCR2 receptors. IL-8 expression is often upregulated in tumors and contributes to angiogenesis and metastasis.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for use at 5 µL (0.03 µg) per test. Each test corresponds to staining a 100 µL sample (10⁵–10⁸ cells/test).

Target Information

Interleukin 8 (IL-8, CXCL8) is a 72 amino acid pro-inflammatory chemokine belonging to the CXC family. It binds to receptors IL-8RA and IL-8RB and acts as a chemoattractant and angiogenic factor. Expression is induced by inflammatory stimuli in macrophages and endothelial cells, where IL-8 is stored in Weibel-Palade bodies.

IL-8 contains the ELR motif and signals through CXCR1 and CXCR2. It was previously known as NAP-1, GCP-1, MONAP, and protein 3-10C. The IL-8 gene is part of a chemokine cluster on chromosome 4q. IL-8 is implicated in tumor angiogenesis and growth and in the pathogenesis of bronchiolitis.
