Target Information
The cytochrome P450 (P450) superfamily of enzymes is one of three enzyme systems that metabolize the fatty acid arachidonic acid (AA) into regulators of vascular tone.
P450 enzymes are monooxygenase enzymes that require cofactors such as NADPH and P450 reductase.
There are over 200 known genes encoding P450s. Epoxygenases metabolize AA to epoxyeicosatrienoic acids (EETs), while omega-hydroxylases produce 19- and 20-hydroxyeicosatetraenoic acids (19- and 20-HETE).
In addition to fatty acid metabolism, P450s are involved in drug and toxin metabolism. Cytochrome P450 3A4 is highly expressed in the liver and small intestine and is inducible by barbiturates, glucocorticoids, and rifampicin.
