Target Information

The p38 MAPK cascade regulates various cellular responses to stress, inflammation, and other stimuli. p38 MAPK is inactive in its nonphosphorylated form and becomes rapidly activated by dual phosphorylation of the Thr-Gly-Tyr motif. There are four isoforms of p38 MAPK, differing in tissue expression and interactions with upstream activators and downstream effectors. Upon exposure to tumor necrosis factor, interleukin-1, or heat shock, MAPK kinase-3/6 phosphorylates Thr180 and Tyr182 residues in p38 MAPK. Phospho-p38 MAPK subsequently activates transcription factors such as ATF-2, CHOP-1, and MEF-2 through phosphorylation.
