Parkinson’s disease is the second most common neurodegenerative disorder after Alzheimer’s disease, affecting about 1% of people over 65 and 3% over 75. Mutations in the Park2 gene are the most common known cause of Parkinson’s disease. Although the function of Park2 is not fully understood, it may participate in the ubiquitin-mediated proteolytic pathway. Mutations can cause autosomal recessive juvenile parkinsonism. Alternative splicing produces three known isoforms of undetermined function. Panneuronal expression of Parkin substrate Pael-R induces age-dependent degeneration of dopaminergic neurons in Drosophila, while coexpression of Parkin degrades Pael-R and suppresses its toxicity.
