Product Specific Information

This antibody is purified from rabbit serum by sequential epitope-specific chromatography and contains sufficient material for approximately 10 mini-blots. It has been negatively preadsorbed using a non-phosphopeptide corresponding to the phosphorylation site to remove reactivity with non-phosphorylated IRS-1. The final product is affinity purified using an IRS-1 peptide phosphorylated at serine 616.

The antibody has been validated in Western blotting, and previous lots have also been used in immunohistochemistry. In Western analysis, the positive control was human embryonic kidney 293T cells transfected with a vector encoding IRS-1 and stimulated with phorbol ester (TPA).

Target Information

IRS-1 (Insulin Receptor Substrate 1) is a major substrate of the insulin receptor and is phosphorylated in response to insulin, IGF-1, and IL-4 stimulation. Phosphorylation occurs on serine, threonine, and tyrosine residues across various tissues. Casein kinase II mediates insulin-stimulated phosphorylation of IRS-1 at several sites, including Thr502 and Ser99. IRS-1 also contains three putative 14-3-3 binding sites (Ser270, Ser374, Ser641), with Ser270 located within the phosphotyrosine-binding domain responsible for insulin receptor interaction.
