Product Specific Information

The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular signaling leading to cellular activation and proliferation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤1.0 µg/test (100 µL final volume). Optimal performance should be determined empirically.

Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is designed for cytometers equipped with a 355 nm ultraviolet laser. Ensure instrument compatibility before use.

When using multiple polymer dye-conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR complex surface expression. Activation through the TCR involves CD3 gamma, delta, epsilon, and zeta subunits, which are structurally related members of the immunoglobulin superfamily encoded on chromosome 11.

CD3 subunits have long cytoplasmic tails that associate with signal transduction molecules via double tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Defects in the CD3 gene are associated with CD3 immunodeficiency.
