Target Information
CYLD is a 956 amino acid cytoplasmic deubiquitinating enzyme belonging to the ubiquitin carboxy-terminal hydrolases (UCH) family. It contains three CAP-GLY domains, a proline-rich region, an SH3 binding domain, and a catalytic domain homologous to UCH. CYLD functions as a tumor suppressor, negatively regulating TRAF2 and NF-κB signaling pathways, and plays a role in the IκB kinase pathway. It exhibits deubiquitinating activity toward non-Lys-48-linked polyubiquitin chains, with TRAP1 identified as a substrate. Mutations in CYLD are associated with cylindromatosis, multiple familial trichoepithelioma, and Brooke-Spiegler syndrome.
